Design Notes

The Gli1 zinc finger sequence (Part:BBa_K165007) (with a previously added linker sequence) was ligated into an existing BioBrick vector containing the Elowitz RBS site (Part:BBa_B0034). The vioC gene was obtained by PCR from an existing BioBrick of violacein operon (Part:BBa_K274002) and ligated as a back insert into the RBS_zinc-finger construct.

The vioC sequence was obtained by PCR from an existing part, which was designed by the Cambridge 2009 iGEM team (Part:BBa_K274002). The Gli1 zinc finger was also obtained from the Registry of parts (Part:BBa_K165007).
